The tty should be in /dev not /sys

thumbDV is connected to ttyUSB0.

Ko,

I don't have a Zero to test on -- however, this is a simple piece of hardware and software.

There are test python scripts in https://github.com/dl5di/OpenDV/tree/master/DummyRepeater/DV3000 to make sure you are able to talk to the ThumbDV on /dev/ttyUSBx  (x being the number for the port).  Look at the script to see what settings you need.

Next run the ambeserver from the command line

sudo ambeserver -p /dev/ttyUSB0 -s 460800  # As an example

Watch for errors and from a second terminal run the ambesocketest.py https://nw-digital-radio.groups.io/g/ambe/wiki/AMBEserver-Addendum#Testing-the-AMBEserver

If that all works, then move on to debugging why your systemctl file isn't working.

Make sure nothing else is trying to talk to the port at the same time when running tests or ambeserver.
